Construction and use of a dialysis chamber for investigating in situ the toxicity of heavy metals on bacteria. ArchiMer

Teflon dialysis sampling chambers have been employed to study the toxicity of ZnCl sub(2) on a cadmium-sensitive bacterium, Acinetobacter) CS 13, in an aquarium with running seawater. A concentration of 10 mg Zn super(++) l super(-1) did not influence the cell densities in the chambers, but cell densities decreased at zinc concentrations of 15 and 20 mg l super(-1). In seawater with 20 mg Zn super(++) lu-1 the bacteria accumulated zinc from 827 to 2813 mu g g super(-1) cells dry wt after 1 d. After 2 d the zinc concentration increased to 4625 mu g g super(-1) dry wt. Ce travail a pour objectif de déterminer le niveau de contamination des produits halieutiques mauritaniens. Pour cela, nous avons étudié les polluants chimiques (Cu, Fe, Zn, Cd et Hg) dans des moules (<Perna perna>) prélevées au niveau de la baie du Lévrier en Mauritanie sur une période de quatre mois (novembre 1998 à février 1999). Sur les 400 échantillons analysés les moyennes de concentrations trouvées restent faibles par rapports à celles trouvées dans d'autres zones peu polluées (Amiard et al. 1986b). Zinc and water intake in rats: investigation of adrenergic and opiatergic central mechanisms BJMBR

We have demonstrated that central administration of zinc in minute amounts induces a significant antidipsogenic action in dehydrated rats as well as in rats under central cholinergic and angiotensinergic stimulation. Here we show that acute third ventricle injections of zinc also block water intake induced by central ß-adrenergic stimulation in Wistar rats (190-250 g). Central inhibition of opioid pathways by naloxone reverses the zinc-induced antidipsogenic effect in dehydrated rats. After 120 min, rats receiving third ventricle injections of isoproterenol (160 nmol/rat) exhibited a significant increase in water intake (5.78 ± 0.54 ml/100 g body weight) compared to saline-treated controls (0.15 ± 0.07 ml/100 g body weight). In the present study we characterized the capacity of zinc to protect lipids and proteins from Fe2+-initiated oxidative damage. The effects of zinc on lipid oxidation were investigated in liposomes composed of brain phosphatidylcholine (PC) and phosphatidylserine (PS) at a molar relationship of 60:40 (PC:PS, 60:40). Lipid oxidation was evaluated as the oxidation of cis-parinaric acid or as the formation of 2-thiobarbituric acid-reactive substances (TBARS). Zinc protected liposomes from Fe2+ (2.5-50 muM)-supported lipid oxidation. Relations among Valencia orange yields with soil and leaf nutrients in Northwestern Paraná, Brazil BABT

The Valencia orange orchards established on soils of low fertility in the Northwest region of Paraná State, Brazil, have showed symptoms of Mg deficiency and reduced fruit yields. The objective of this study was to verify the relationship between yield with soil and leaf nutrients during 1996/97 growing season. Two sites of low and high productivity were selected in seven orchards. Leaf and soil samples (fertilized rows and interrows) were collected in 1996. The results showed that the citrus yields were negatively related with soil Mg/K and Ca+Mg/K ratios in the fertilized rows, and fruit weight positively correlated with leaf Zn in the low productivity orchards. Amorphous phosphate granules are present in vertebrate and invertebrate organisms. The functions attributed to these structures depend on their mineral contents and organic matrix composition. In the present study we have determined zinc concentrations in the hepatopancreas of the crab Ucides cordatus from regions contaminated with zinc, and the elemental composition of hepatopancreal phosphate granules. Differential effects of lead and zinc on inhibitory avoidance learning in mice BJMBR

We studied the effects of chronic intoxication with the heavy metals lead (Pb2+) and zinc (Zn2+) on memory formation in mice. Animals were intoxicated through drinking water during the pre- and postnatal periods and then tested in the step-through inhibitory avoidance memory task. Chronic postnatal intoxication with Pb2+ did not change the step-through latency values recorded during the 4 weeks of the test (ANOVA, P>0.05). The medial cerebral cortex of lizards, an area homologous to the hippocampal fascia dentata, shows delayed postnatal neurogenesis, i.e., cells in the medial cortex ependyma proliferate and give rise to immature neurons, which migrate to the cell layer. There, recruited neurons differentiate and give rise to zinc containing axons directed to the rest of cortical areas, thus resulting in a continuous growth of the medial cortex and its zinc-enriched axonal projection. Zinc detoxification by a cyanobacterium from a metal contaminated bay in Brazil BABT

We describe here the trapping of zinc in polyphosphate granules of the cyanobacterium Synechocystis aquatilis NPBS-3. Cells were cultured in 25 µM of zinc chloride and prepared for electron microscopy and energy-dispersive X-ray analysis. Some ultrastructural features were changed by zinc exposure, the increase of glycogen granules number being the main change. The polyphosphate granules contained phosphorus, sulphur, calcium, iron and zinc. The trapping of zinc in polyphosphate granules seemed to be an effective way of detoxifying the metal and surviving in the bay. As a non-specific mechanism, these polyphosphate granules could also be effective in trapping other metals in excess.

Manejo de hábitat y microorganismos para degradar efluentes industriales: un estudio de caso Ecología austral

Se evaluó la eficiencia de prácticas de manejo de hábitat y microorganismos para degradar efluentes en una industria metalúrgica de Córdoba, Argentina. Para optimizar la actividad de organismos del agua, el suelo y la rizosfera de las plantas, se manejaron tres hábitats: acuático, de humedales y agrícola. Las prácticas fueron: introducción de vegetación palustre y terrestre, aireación del agua, laboreos, remoción de sedimentos e inoculación con bacterias seleccionadas. Se evaluaron los siguientes parámetros: pH y contenido de bacterias en el agua, y contenido de aceites, Cr y Zn en los sedimentos, suelo y napa freática. Desde el punto de vista nutricional es necesario conocer la concentración sérica de algunos minerales en niños y para poder interpretar los resultados es imprescindible comparar con valores de referencia. El objetivo de este trabajo es determinar valores de referencia de cobre, zinc y selenio en suero de niños. Se utilizaron muestras de suero de niños de ambos sexos (n = 42) clínicamente sanos, elegidos al azar, entre 1 mes y 12 años de edad, concurrentes para control al Servicio de Pediatría del Hospital de Clínicas "José de San Martín", Ciudad Autónoma de Buenos Aires. Correlation between heavy metal ions (copper, zinc, lead) concentrations and root length of Allium cepa L. in polluted river water BABT

The present work was performed using the common onion (Allium cepa L.) as a bioindicator of toxicity of heavy metals in river water. The test waters were collected at two sampling sites: at the beginning and the end of the Toledo River. The bulbs of A. cepa L. were grown in test water with nine concentration levels of copper, zinc and lead from 0.1 to 50 ppm. In the laboratory, the influence of these test liquids on the root growth was examined during five days. For test liquids containing below 0.03-ppm dissolved Cu the root growth was reduced by 40% However, the same reduction occurred for 1-ppm dissolved Zn. For dissolved Pb, results reveal toxicity above 0.1 and 0.6 ppm at the beginning and the end of the Toledo river water, respectively.

Neo-Timm staining in the thalamus of chronically epileptic rats BJMBR

The thalamus is an important modulator of seizures and is severely affected in cholinergic models of epilepsy. In the present study, chronically epileptic rats had their brains processed for neo-Timm and acetylcholinesterase two months after the induction of status epilepticus with pilocarpine. Both controls and pilocarpine-treated animals presented neo-Timm staining in the anterodorsal nucleus, laterodorsal nucleus, reticular nucleus, most intralaminar nuclei, nucleus reuniens, and rhomboid nucleus of the thalamus, as well as in the zona incerta. The main aim of this paper was to investigate the effect of chromium (Cr), copper (Cu) and zinc (Zn) on nitrate reductase (NR) activity and oxidative stress responses in the moss Polytrichum commune. Cr, Cu and Zn resulted in the inhibition of NR activity. A decline in total chlorophyll content was observed after 24 and 48 h of metal treatment. Accumulation of the metals showed a dose and time dependent increase. High accumulation of Cu, Cr and Zn were seen in moss shoots after 24 and 48 h of treatment. Treatment of Cr, Cu and Zn for 24 or 48 h resulted in the increase of malondialdehyde (MDA) content in moss shoots.
